V-Star 250 runs on one cyl.
I've got a 2009 V-Star 250 that has been sitting for a few weeks. When I start it up, only the front cylinder is firing. I know the rear cylinder is getting spark, so I assume it's due to lack of fuel. It looks like the two cylinders share the same carb and intake manifold, so what could be keeping the gas from getting to the rear cylinder?

On a single carb system, when one cylinder quits firing, the first thing you should do is replace the plugs.
Yes, even though you did a simple test to see if the plugs were firing.
Sometimes those tests can lie if not done exactly right.
While not totally impossible, it is highly unlikely that only one cylinder has a fuel problem.

I did swap the plugs, but it made no difference. Still only runs on front cylinder.

OK, assuming that you aren't overlooking something really easy and obvious............
probably the next thing you should do is a compression check.
Long shot that you have a valve problem.
If it has the typical double-fire, series wired, single coil arrangement for the plugs, then there isn't much that will cause only one plug to misfire........except a shorted plug.
AND with a single carb, there isn't much that will cause only one cylinder to have a bad mixture......except a bad vacume leak or a stuck/warped valve........or a piston with a hole in it !!!
